Assembly of a patchy protein into variable 2D lattices via tunable multiscale interactions

Self-assembly of molecular building blocks into higher-order structures is exploited in living systems to create functional complexity and represents a powerful strategy for constructing new materials. As nanoscale building blocks, proteins offer unique advantages, including monodispersity and atomically tunable interactions. Yet, control of protein self-assembly has been limited compared to inorganic or polymeric nanoparticles, which lack such attributes. Here, we report modular self-assembly of an engineered protein into four physicochemically distinct, precisely patterned 2D crystals via control of four classes of interactions spanning Ångström to several-nanometer length scales. We relate the resulting structures to the underlying free-energy landscape by combining in-situ atomic force microscopy observations of assembly with thermodynamic analyses of protein-protein and -surface interactions. Our results demonstrate rich phase behavior obtainable from a single, highly patchy protein when interactions acting over multiple length scales are exploited and predict unusual bulk-scale properties for protein-based materials that ensue from such control.

Building two-dimensional materials one row at a time: Avoiding the nucleation barrier

No barriers to growing a row Classical nucleation theory predicts that two-dimensional islands on a surface must reach a critical size before they continue to grow; below that size, they dissolve. Chen et al. used phage display to select for short peptides that would bind to molybdenum disulfide (MoS 2 ) (see the Perspective by Kahr and Ward). Hexagonal arrays of these peptides grew epitaxially as dimers but without a size barrier—the critical nuclei size was zero. Although two-dimensional arrays formed, growth occurred one row at time. In vitro single-cell dissection revealing the interior structure of cable bacteria

Significance Cable bacteria were recently discovered as the facilitators of electron transfer over centimeter distances in marine sediments. In this work, we explore the unique structure of the cable bacteria, using atomic force microscopy-based single-cell in vitro dissection. We identify different types of bacterial cell–cell junctions and continuous dimeric strings hidden under the outer membrane that pass through the cell junctions. The strings seem to serve an important function in maintaining the integrity of individual cable bacteria cells as a united filament. On the basis of our findings, we propose a model for the division and growth of the cable bacteria, which illustrate the possible structural requirements for the formation of centimeter-length filaments in the recently discovered cable bacteria.
